Project group 1 (led by Jonathan Blackburn): Development of standardised array platform
One or two array platforms that have the following characteristics:
- can be used for both synthetic and cell or tissue derived oligosaccharides
- optimised presentation of carbohydrate to carbohydrate-binding proteins (CBPs)
- both carbohydrate arrays and the CBP binding can be analysed by mass spectrometry, SPR and fluorescence
|
Project group 3 (led by Ten Feizi and Jerry Turnbull): Preparation and screening of natural arrays
- Establishment of arrays of diverse sequence-defined natural oligosaccharides and those from glycomes of cells and tissues for defining the saccharide ligands of CBPs
- Evaluation of the arrays for identifying the ligands for CBPs using recombinant soluble and cell-associated Siglecs, and
heparin-binding proteins, as model systems, and extending to other members of the immunoglobulin super family
- Validation of arrays in comparison to other conventional binding methods
